     This product can be used for qualitatively detect p16 and Ki-67 proteins in cells in vitro. Immunocytochemistry is a method for the localization or qualitative detection of intracellular antigens (test targets) by using the principle of specific binding of antigens to antibodies and the coloration of the labeled antibodies through a chemical reaction.
      The primary antibodies for this product consist of a mixture of mouse monoclonal anti-human p16 antibody and rabbit monoclonal anti-human Ki-67 antibody. Corresponding secondary antibodies are a mixture of alkaline phosphatase (AP)-conjugated goat anti-mouse IgG antibody and horseradish peroxidase (HRP)-conjugated goat anti-rabbit IgG antibody. When AP reacts with the NF chromogen solution, a red coloration can be observed, whereas when HRP reacts with the DAB chromogen solution, a brown coloration will result. The cellular localization and expression levels of p16 and Ki-67 proteins are inferred by observing the coloration of the cells under a microscope.
The red coloration in this product indicates the presence of the p16 antigen, which is localized primarily in the cytoplasm of cells, occasionally in the cell nucleus. The brown coloration, on the other hand, indicates the presence of the Ki-67 antigen, which is localized within the cell nucleus.
